胰腺癌PC 3细 胞 【Abstract】 Objective To prepare 5 fluorouracil magnetic albumin microspheres (5 FU MAMS) and discuss its inhibitive action combined with constant external magnetic fields on human pancreatic carcinoma PC 3 cells in vitro. Methods 5 FU MAMS was prepared by emulsion heat stabilization technique and their physico chemical property detected. The inhibition rates (IR) of 5 FU MAMS combined with constant external magnetic fields were assayed by using MTT assay.Results The mean diameter of 5 FU MAMS was 200 1000 nm, with round shape, good magnetic response and 5 FU loading of 5%. The tumor growth showed no difference on tumor cells in both pure magnet group and MAMS group. The IR of 5 FU MAMS and 5 FU at same drug concentration was similar, with insignificant statistical difference (P0.05). Compared with pure 5 FU MAMS and free 5 FU, IR was significantly increased after 5 FU MAMS was combined with constant external magnetic fields (P0.05). Conclusions MAMS is a safe drug carrier. The preparation method and changes of dosage form exert no action on anticancer activity of 5 FU. The inhibitive action is significantly enhanced after5 FU MAMS is combined with constant external magnetic fields. 【Key words】 5 fluorouracil magnetic albumin microspheres;

Pancreatic carcinoma PC 3 cells 胰腺癌早期诊断困难,具有病程短、进展快、病死率高的特点, 中位生 存期约6个月, 总体5年生存率低于5%[1]。目前对胰腺癌仍采用手术切除联 合化疗的综合治疗方案。近年来,运用抗癌药物磁性纳米微球联合磁场治疗肿瘤 取得可喜进展。本实验制备5 氟尿嘧啶磁性白蛋白微球(5 fluorouracil magnetic albumin microspheres, 5 FU MAMS)并检测其理化性质,分析其联合恒定磁 场对人胰腺癌PC 3细胞的生长抑制作用,探讨联合磁场后5 FU MAMS抑瘤 作用增强的机制。
1 材料与方法 1.1 材料 5 FU,纯度99%,购自比利时Acros公司;
牛血清白蛋白购自美国Sigma 公司;
超声乳化仪购自江苏金坛医疗仪器厂;
SSX 550型扫描电子显微镜购自 日本;
752型紫外可见分光光度计、Multiskan MK3型酶标仪购自上海分析仪器总 厂;
恒定磁场由北京新海空科技开发中心设计提供;
胰腺癌PC 3细胞系由解放 军302医院肝胆研究中心赠送。
IR=[(对照组OD值-空白调0孔OD值)-(实验组OD值-空白调0孔OD值)/(对 照组OD值-空白调0孔OD值)。IR50%时表示该药物敏感。
1.3 统计学分析 采用SPSS 10.0软件包进行统计学处理,数据以±s表示,组间差异进行t检 验。P0.05为差异有统计学意义。
2 结果 2.1 5 FU MAMS的理化性质检测 2.2 胰腺癌PC 3细胞生长抑制率的检测 3 讨论从本实验结果可知,单纯白蛋白微球和磁场体外对胰腺癌PC 3细胞生长 无影响,白蛋白微球可以作为肿瘤化疗药5 FU的安全载体,载药MAMS和5 FU 有相似的抑瘤效应;
5 FU MAMS联合磁场能明显提高化疗药物的抑瘤效果。
其机理可能是由于磁场对细胞膜有穿透作用,肿瘤细胞在磁场作用下,膜表面的 带电离子由于受到洛仑兹力影响,其运动轨迹被约束,导致了肿瘤细胞膜上的电 子和离子不能正常运动和传递,引起了肿瘤细胞膜微观结构的变化,从而影响了 细胞膜的通透性和有序性 [5]。这就为化疗药物分子更快、更多穿透细胞膜提 供了条件,这可能是磁场与磁性化疗药物协同作用提高其抑制肿瘤细胞生长作用 的原因之1。
